a) therapeutic goal: to increase the number of activated and resting lymphocytes. these observations bring to light that quiescent lymphocytes in aging t cells are activated by pak, which is a putative gtpase activator. selective pak activation leads to a cell type-specific and time-dependent morphological transformation and activation of an accompanying increase in cellular metabolic activity and oxygen consumption, whereas nonselective activation of pak leads to decreased cell viability. the differences in pak activation lead to the conclusion that pak1 is a potential target for treating immunosenescence and that selective pak1 activation may be a new approach to the treatment of immunodeficiency. the authors also showed that this is a novel pathway in p53-dependent apoptosis regulation and pak1 inhibition protects from dna-damage-induced senescence and prevents an age-dependent decrease in the number of activated t cells.
